The Owls slipped to a second successive defeat on Saturday morning, going down 2-1 against Sheffield United at Shirecliffe.
It was Wednesday’s first outing since defeat in the fifth round of the FA Youth Cup away at Blackburn in late January.
Midfielder Rio Shipston was dismissed just after the hour mark for picking up two yellows as the derby honours went to United.
Sydie Peck gave the Blades a half time lead with the midfielder heading home for the hosts three minutes before the break.
Mackenzie Maltby levelled for the Owls four minutes after the restart, finishing inside the box from a Caelan Kilheeney free kick.
But Wednesday were forced to see out the final third of the game with ten men as Shipston received his marching orders after picking up a second yellow card.
And the Blades made their numerical advantage count when Fernando Macedo fired an unstoppable half volley from 20 yards into the bottom corner to seal victory in the derby.
